Biontin AviTag Vectors

N-terminal Biotin AviTag Vectors

The pAN series of vectors are used to express N-terminal AviTag-protein fusions. In April of 1999, Avidity improved the N-terminal AviTag vectors to include a glutamic acid residue which has been shown to greatly enhance biotinylation rates of the AviTag. (Beckett et al., 1999)The AviTag peptide sequence used is the #85 sequence described in Schatz (1993). Three variants of the vector are available to allow cloning into any reading frame.Expression of the AviTagged gene is under the control of the tightly repressed Trc promoter and is IPTG-inducible.

Plasmids pAN-4,5&6

In pAN-4, the ß-lactamase gene (amp) covers bp 873-1733 and lacIq covers 3082-4164. To obtain the numbering for pAN-5 and pAN-6, these values should be incremented by 1 and 2 respectively.

C-terminal Biotin AviTag Vectors

The pAC series of vectors are used to express C-terminal AviTag-protein fusions.
