HEK293细胞中共转染miR-125a 抑制剂表达质粒 (GeneCopoeia HmiR-AN0094-AM01) 、miR-125a 前体表达质粒(GeneCopoeia HmiR0309-MR03) 和miR-125a的靶标克隆质粒(GeneCopoeia HmiT019205-MT02:将LIN28 的3’UTR克隆到Gaussia荧光素酶-碱性磷酸酶双报告载体)。转染24小时后检测报告基因Gaussia 荧光素酶和参照基因碱性磷酸酶的活性。只转染Gluc-LIN28-3’-UTR质粒的细胞中Gaussia 荧光素酶与碱性磷酸酶活性比值设为1 (左数第一条柱),用来标准化其他细胞样品中的报告基因活性。结果表明miR-125a抑制了Gluc-LIN28-3’-UTR中Gluc 70%以上的活性(左数第二条柱)。这种抑制效应可被导入的不同剂量的miR-125a抑制剂表达质粒特异性的阻遏。转染100 ng 抑制剂表达质粒时,细胞内报告基因/参照基因活性比值(右数第一条)已超出单独转染Gluc-LIN28-3’-UTR报告质粒的对照组。这说明miR-125a抑制剂可以阻遏外源和内源性的miR-125a对靶标基因的抑制,提高GLuc-LIN28-3’-UTR转录本的翻译水平。
